This Warhammer 40,000: Space Marine 'behind the scenes' video -- made available by the good guys at Machinima -- explains what Space Marines and Titans are, and why Orks are your main enemy.
What Orks, you ask? WeirdBoyz, Gretchen, Nobz are definitely in. While they aren't mentioned specifically, Shoota Boyz will probably make an appearance too. Because, you know, they shoot and this is a third-person shooter game with melee attacks. Also Chaos.
Space Marine certainly looks better than back when it was first announced, so here's hoping that Relic can make as fine a 40k action game as they make real-time strategy games.

Looking good! I am psyched for this. I am spoiled by DoW II: Retribution though and want to play as the other races. I love the Orks... it's a shame I will have to reduce them to so many giblets.
@Occams Yeah, they've got Ork Gargants. Though I can see a real enterprising Ork boss rallying his boys to steal a titan and make it into whatever he wants.
@ Dv8thwonder: Most games featuring anything like a space marine is considered generic because it's basically a rip of Warhammer 40K. This is the source material for everything from Gears to Starcraft. No idea if the actual game will be better than Gears or not, but don't knock it for being so badass everyone else wanted to copy it.
I don't know if the game still has them but Eldar and the Imperium used to have 30 ft tall mini-Titans called Knights that were even smaller than Warhound titans. Orcs had something similar called Stompa's that were built by those lower ranking Mekboyz who couldn't manage a gargant but still wanted to contribute something cool to a Waaaagh.
A Space Marine's cover is his faithful devotion to his duty to the Emperor of mankind. It is his righteous hatred of the heretic, the xeno, the rebel, and the daemon. It is his phenomenal healing abilities, his wargear, and his power-armor. It is his knowledge that his brothers will recover the geneseed from his body and return it to the chapter should he fall in battle.
Also a high toughness and rolling under the armor save threshold helps a whole lot.
My only real concern with this is whether they'll be able to pull off the feel of weight of a SPEHS MEHREEN and their weapons. Gears managed it by ploughing the player into cover like they were trying to knock it down, but these guys don't really care for chesthigh walls (not to mention that 'chest-high' to a SM is about 6 foot, those guys are HUGE).
More than that though, are the default weapons. Laser guns are pretty standard pew-pew fare of different sizes, and even plasma guns can probably be made to look suitably awesome with some fancy particle effects and hissing noises. But the bog-standard, absolute bare-minimum weapon a SM ever has is a pistol capable of firing miniature rocket-propelled grenades.
I want you to think about that for a second. It's a pistol the size of your chest, and it fires ROCKET PROPELLED GRENADES. I don't know how they're gonna be able to justify more than one shot kills on your standard orky types.
