Picross doesn't really scream out at you if you've never tried it before; it just looks like a grid with numbers. But it's one of Nintendo's best puzzle games, with a lot of that "Nintendo" charm: sound effects, cool references to classic games, super addictive. My wife and I were hooked on this for months.
Dragon Quest Heroes: Rocket Slime is a sublimely happy game. Adorable adventure game with some deep strategy mixed in.
Also Space Invaders Extreme is surprisingly good. It's put out by Square-Enix and I promise you, it's not just another rehash of Space Invaders. I thought to myself, I've played Space Invaders a thousand times. I don't need an update with "edgy" graphics. But I was so wrong. This game is like Space Invaders mixed with Rez and with Aphex Twin doing the soundtrack. There's a cool symmetry that goes on with the music and the way you fire the missiles. It feels like you're creating songs as you play. I tried it begrudgingly and ended up loving it.

Pick up an R4 unless its against your beliefs. It will save you so much money because you get to test out games that look good but are utter crap. If you do get an R4 you can always buy a game if you want to support the industry. Regarding you post I agree with TWEWY it is FUCKING amazing and one of my favorite games of all time, Tetris DS is a favorite of mine, And Space Invaders is really good as well. I never thought I really liked tetris but now I am addicted and it is my most played game.
If you get Animal Crossing, change the DS's internal clock to day time during your night shift. Nothing happens at night in Animal Crossing.
If you like import games, make sure to pick up Ouendan 2,ts the Japanese version of Elite Beat Agents, and it is soooo much better. The game starts out really easy, but as you progress through the songs, they get harder, and harder, until you have to try a song tens of times to beat it, but it never gets frustrating. The game also has amazing music, if you want to know more about Japanese music, play this game. But you have to pay like $65 bucks for it.
Rhythm Tengoku Gold is suposed to be coming to NA sometime as "Rhythm Heaven". It's a rhythm based minigame collecion developed by the guys who made Wario-Ware. I haven't played it yet, but it is supposed to be really good.
Kirby: Canvas Curse is really good. Kirby takes to form of a ball, and you have to draw little rainbow lines that Kirby follows. It came out like right after the DS was released, but it is one of the early cases of touch screen gameplay done right.
Pretty much any of the Castlevania games on DS are really great. I like Dawn of Sorrow (the first one) the best. I haven't played Order of Ecclisia yet, but it is supposed to be really good too.
Tetris DS, in my opinion, is the best version of Tetris out there. It has tonnes of different game modes, great online multiplayer, and lots of great throwbacks toward Nintendo games of old. There really is nothing better than listening to remixes of Nintendo songs while playing Tetris.
Also, Zelda Minish Cap is a great Zelda game. (Ya, I know, I know, it's not a DS game, but you can play it on your DS :P) It is actually my favourite handheld Zelda game of all time, even though it's a Capcom game.

You can't go wrong with the three Castlevania DS games if you're into the series. Most metroidvania games (as with the GBA ones) are well worth playing.
Elite Beat Agents is dirt cheap now, under 10 bucks if you look around. I loved it and I hate rhythm games.
Also, if you're into puzzle games opt for Tetris DS and Planet Puzzle League (aka Panel de Pon/Tetris Attack/Pokemon Puzzle League) over the Brain Training games for more replayability.
Can't forget Mario Kart DS. The online is probably is lacking now but I'd still consider it the best MK game.
Other than that, if you're into RPGs the two Golden Sun GBA games are very good. Have fun finding those though.

As Triestero said, Space Invaders extreme is great. Its cheap and really fun.
Geometry Wars Galaxies is quite good for Geometry Wars on the DS. Not sure if you enjoy Geometry Wars but it works surprisingly well using the d-pad and stylus. There are a ton of levels plus retro evolved which is sharable.
N+ is N+ on the DS. If you enjoyed it on XBLA, chances are you will like it in portable form. Its $20 and works well as a pick up and play game.
As far as GBA games go I would recommend Minish Cap, Mario & Luigi Superstar Saga and Rhythmn Tengoku which is import only. Its from the people that made Warioware and its crazier than that. Also Warioware on the DS is really fun if you can find it.
